Truelab™ workstation is one such platform meeting all these requirements. Truelab™ Uno / Truelab™ Uno Dx / Truelab™ Duo / Truelab™ Quattro is a hand held, battery operated, easy to carry nucleic acid amplification workstation consisting of lightweight, portable, mains / battery operated Truelab™ Uno Dx / Truelab™ Duo / Truelab™ Quattro Real Time micro PCR Analyzer, TrueprepTM AUTO Sample Prep Device, room temperature stable Truenat™ micro PCR chips and TrueprepTM AUTO Sample Prep kits. It is a portable system and results are reported in less than an hour. It is enables accurate and specific disease diagnosis at Point-of-care PoC or near-care settings, and is suited especially for resource limited settings. Even the peripheral laboratories with minimal infrastructure and minimally trained technician can easily perform these tests routinely in their facilities and report PCR results in less than an hour. The device works on the principle of real time Polymerase Chain Reaction PCR . The device uses Truenat™ which is a disposable, microchip made of LTCC which contains pre-loaded and room temperature stabilized PCR reagents. Only the purified nucleic acid sample should be added to start the test. The signal is captured by the microPCR device using an opto-electronic circuit and the results displayed on the screen as amplification curves. The device has fast turn around time with ~35 minutes to complete both amplification and detection. The Tuelab™ Uno / Truelab™ Uno Dx / Truelab™ Duo / Truelab™ Quattro micro-PCR has automated disease surveillance capability, wherein the results from a diagnostic run would be automatically sent to a central server in encrypted form , for analysis and review by health care authorities. Trueprep Auto a sample prep System is an electromechanical system pre-programmed to sequentially heat, mix and add reagents to the contents of the cartridge placed in the cartridge holder and has the two line LCD screen that displays the status. Truelab Uno Uno / Truelab™ Uno Dx / Truelab™ Duo / Truelab™ Quattro is a Real time PCR based system. It involves in amplifying and detecting the target DNA during every cycle, thus generating accurate information for the presence of the target DNA and its quantity in each reaction. The principle of Real Time Reverse Transcription Polymerase Chain Reaction real time RTPCR based on Taqman chemistry. The patient sample is first extracted using Trueprep™ AUTO Sample Prep Device and Trueprep™ AUTO sample prep kit and six 6 µL of the purified RNA/DNA is then dispensed into a microtube containing freeze dried PCR reagents, including reverse transcriptase RT . After allowing approximately 20 seconds for the dried PCR reagents to get hydrated with the RNA/DNAsample, the entire contents is pipetted out and dispensed into the reaction well of the Truenat™ chip. The Truenat™ chip is then inserted in the Truelab™ Real Time micro PCR Analyzer where the RNA/DNA is first converted into complementary DNA cDNA by the RT enzyme and further thermal cycling takes place. A positive amplification causes the dual labeled fluorescent probe in the Truenat™ chip to release the fluorophore in an exponential manner which is then captured by the built-in opto-electronic sensor and displayed as amplification curve on the analyzer screen, on a real time basis during the test run. The Cycle threshold Ct is defined as the number of amplification cycles required for the fluorescent signal to cross the threshold i.e. exceed the background signal . Ct levels are inversely proportional to the amount of target nucleic acid in the sample. i.e the lower the Ct level the greater is the amount of target nucleic acid in the sample . In the case of negative samples, amplification does not occur and a horizontal amplification curve is displayed on the screen during the test run. Trueprep Auto is proprietary matrix based extraction. It is fully automatic. It works on proprietary firmware. Truelab Uno Dx is Real time PCR. It is a portable,handheld system.
